However,\u00a0several neurodegenerative diseases present dysregulation of iron homeostasis\u00a0derived from mitochondrial dysfunction, inflammatory conditions, decreased\u00a0glutathione levels, and oxidative damage, resulting in downstream protein aggregation,\u00a0lipid peroxidation, and nucleic acid modification. In this chapter, the\u00a0mechanisms by which iron homeostasis is lost in Parkinson\u2019s disease (PD) are\u00a0discussed. The relevance of endogenous toxins such as mediators of mitochondrial\u00a0dysfunction, the relationship between inflammation and iron\u00a0dyshomeostasis, and the role of hepcidin as a neuroprotective agent are also\u00a0addressed. A model is proposed that involves a positive feedback loop between\u00a0mitochondrial dysfunction, inflammation, and increased iron content in dopaminergic\u00a0neurons, which, if unchecked, ends in substantia nigra (SN) neuronal\u00a0death.<\/p>\n","protected":false},"featured_media":964,"template":"","class_list":["post-1393","publicacion","type-publicacion","status-publish","has-post-thumbnail","hentry","revista-handbook-of-neurotoxicity"],"acf":[],"_links":{"self":[{"href":"https:\/\/cib.udp.cl\/en\/wp-json\/wp\/v2\/publicacion\/1393","targetHints":{"allow":["GET"]}}],"collection":[{"href":"https:\/\/cib.udp.cl\/en\/wp-json\/wp\/v2\/publicacion"}],"about":[{"href":"https:\/\/cib.udp.cl\/en\/wp-json\/wp\/v2\/types\/publicacion"}],"wp:featuredmedia":[{"embeddable":true,"href":"https:\/\/cib.udp.cl\/en\/wp-json\/wp\/v2\/media\/964"}],"wp:attachment":[{"href":"https:\/\/cib.udp.cl\/en\/wp-json\/wp\/v2\/media?parent=1393"}],"curies":[{"name":"wp","href":"https:\/\/api.w.org\/{rel}","templated":true}]}}
